What version of tikiwiki are you running? This sounds like a old Windows login bug where http://localhost doesnt work, but if you move tikiwiki to a subfolder http://localhost/tikiwiki it works.
The perm denied message is just the internal tikiwiki permissions system. Take a look at the TikiMovies for Premilinary settings to get your permissions setup.
